Biofilm Overview • Heterogeneous – Differing oxygen and nutrient gradients • Grow slowly • Unique phenotype – Different to planktonic state – Conversion of : • Non mucoid phenotype mucoid • Linked to over production of alginate and exopolysaccharide

Stages of Biofilm formation 1. Bacterial attachment 2. Irreversible adhesion 3. Microcolony development 4. Maturation 5. Dispersion

Bacterial attachment & Irreversible adhesion • LecB virulence factor – decreases ciliary beat of airway epithelial cells • Flagella and Twitching important in attachment • Psl, Pel and alginate polysaccharide required – Composed of: • Mannose, galactose, rhamnose and xylose • Rhl QS system important at all stages

Maturation & Dispersion • Formation of water and salt channels • Extracellular polymeric substance – ill-defined mix of: • Polysaccharides • Nucleic Acids • Proteins • Phage lyse bacteria to begin process of dispersion • These dispersed bacteria have reverted to planktonic state • Small colony variants (SCV) constitute a slow-growing subpopulation of bacteria with distinctive phenotypic and pathogenic traits. Phenotypically, small colony variants have a slow growth rate, atypical colony morphology and unusual biochemical characteristics, making them a challenge for clinical microbiologists to identify.

Quorum Sensing • Mutants lacking Las1 gene form poor biofilm structures • QS Mutants more susceptible to antibiotics and phagocytosis • QS regulation of catalase and superoxide dismutase important in H2O2 resistance
